Mycoplasma detection by the direct culture procedure is complicated, time-consuming (i.e. about five weeks) as well as very difficult due to the fact that some Mycoplasma are more elusive to detect than others. PCR has been shown to be highly sensitive, specific and a rapid method for the detection of Mycoplasma contamination in cell cultures.

Application
The system is set up for the detection of various mycoplasma species contaminating cell cultures M.fermentans, M.hyorhinis, M.arginini, M.orale, M.salivarium, M.hominis, M.pulmonis, M.arthritidis, M.bovis, M.pneumoniae, M.pirum and M.capricolum), as well as Acholeplasma and Spiroplasma species, with high sensitivity and specificity. The kit contains positive control (DNA template) and internal control (DNA template) to exclude the possibility for PCR inhibition by the test sample (false negative).
Principle of the method
N-GARDE Mycoplasma PCR Reagent set detects the presence of mycoplasmas in cell culture supernatants by PCR amplification using a reaction mix and a pair of short oligonucleotides hybridizating to a specific sequence on the genome of interest and used as primers for the DNA polymerisation. Cycles of DNA denaturation, primers annealing and DNA polymerisation allow to increase the number of specific sequence from few copies to millions, quantities that can be visualized by agarose gel electrophoresis. After agarose gel electrophoresis, positive samples will generate a 270bp fragment.
